Activation of NF-κB and p300/CBP potentiates cancer chemoimmunotherapy through induction of MHC-I antigen presentation

چکیده
Significance T cells recognize their targets via T-cell receptors (TCRs), which in the case of CD8 + bind to MHC-I:antigen complexes on surface target cells. Many cancer evade immune recognition and killing by down-regulating MHC-I AgPPM. Here, we show how histone acetyl transferases p300/CBP together with NF-κB epigenetically regulate expression molecules, immunoproteasome subunits, peptide transporter enable proper antigen presentation. Notably, this pathway is frequently disrupted human cancers. We now that certain chemotherapeutics can augment presentation activation, thereby enhancing cell effector CTLs.

MAPK kinase 3 potentiates Chlamydia HSP60-induced inflammatory response through distinct activation of NF-κB.
Chlamydia pneumonia (C. pneumonia) remains one of the leading causes of bacterial pneumonia and has been implicated in the pathogenesis of some inflammation-related diseases, such as asthma, chronic obstructive pulmonary disease, and vascular diseases. Heat shock protein 60 is one of the pathogenic components of C. pneumonia that is closely associated with the inflammatory disorders. However, t...

متن کاملImproving vaccine potency through intercellular spreading and enhanced MHC class I presentation of antigen.
The potency of naked DNA vaccines is limited by their inability to amplify and spread in vivo. VP22, a HSV-1 protein, has demonstrated the remarkable property of intercellular transport and may thus provide a unique approach for enhancing vaccine potency. Therefore, we created a novel fusion of VP22 with a model Ag, human papillomavirus type 16 E7, in a DNA vaccine that generated enhanced sprea...
متن کاملMHC class I antigen presentation--recently trimmed and well presented.
Presentation of antigenic peptide to T cells by major histocompatibility complex (MHC) class I molecules is the key to the cellular immune response. Non-self intracellular proteins are processed into short peptides and transported into endoplasmic reticulum (ER) where they are assembled with class I molecules assisted by several chaperone proteins to form trimeric complex. MHC class I complex l...
متن کاملHLA-F and MHC-I open conformers cooperate in a MHC-I antigen cross-presentation pathway.
Peptides that are presented by MHC class I (MHC-I) are processed from two potential sources, as follows: newly synthesized endogenous proteins for direct presentation on the surface of most nucleated cells and exogenous proteins for cross-presentation typically by professional APCs. In this study, we present data that implicate the nonclassical HLA-F and open conformers of MHC-I expressed on ac...
